Just starting Sinemet. How long for effect?

My wife's neurologist ordered several imaging tests to rule out tumors or other spine/brain issues. A different test showed dopamine was about 2 standard deviations below average for my wife's age (60). Her neurologist has started her on Sinemet (Levodopa/Carbidopa), with a dose ramping from a 50/12.5mg once daily, to an end point of 100/25mg 3x daily over a six week period. She's still in week one and after 5 days on the 1x daily 50/12.5mg she's only gotten worse.

Has anyone here on the the forum started Sinemet that could tell me their experience? The one lady I found on YouTube mentioned she felt relief at the 1st dose. Have others felt immediate relief? Has anyone failed to respond to Sinemet at all?

  1. When first diagnosed with a very low dosage of Sinamet I got some very mild relief. However as the dosage was increased I experienced extreme side effects. I am one who cannot tolerate levodopa in any form and am highly drug sensitive. Presently I am on no PD meds at all. I can tolerate the mild PD symptoms and am trying to control them with diet, exercise and rest. I do have another movement disorder and severe arthritis that make it a bit of a challenge but am on other meds for that. Wishing your wife the best in finding what works best for her.     1. Thanks so much for the reply. The doctor suspects my wife has MSA, and response to Levodopa will probably be the final determining factor.
